Loss of the Otx2-Binding Site in the Nanog Promoter Affects the Integrity of Embryonic Stem Cell Subtypes and Specification of Inner Cell Mass-Derived Epiblast

• Otx2 binds to the promoter/enhancer region of Oct4, Sox2, and Nanog in ESCs and EpiSCs • Otx2 binding to the Nanog promoter helps maintain the integrity of ESC compartments • Loss of this Otx2-binding site induces primed-like features in ESCs • Otx2 regulation of Nanog contributes to ICM differentiation of the epiblast Mouse embryonic stem cells (ESCs) and the inner cell mass (ICM)-derived epiblast exhibit naive pluripotency. ESC-derived epiblast stem cells (EpiSCs) and the postimplantation epiblast exhibit primed pluripotency. Although core pluripotency factors are well-characterized, additional regulators, including Otx2, recently have been shown to function during the transition from naive to primed pluripotency. Here we uncover a role for Otx2 in the control of the naive pluripotent state. We analyzed Otx2-binding activity in ESCs and EpiSCs and identified Nanog, Oct4, and Sox2 as direct targets. To unravel the Otx2 transcriptional network, we targeted the strongest Otx2-binding site in the Nanog promoter, finding that this site modulates the size of specific ESC-subtype compartments in cultured cells and promotes Nanog expression in vivo, predisposing ICM differentiation to epiblast. Otx2-mediated Nanog regulation thus contributes to the integrity of the ESC state and cell lineage specification in preimplantation development.

NASA Orders Second Boeing Crew Mission to International Space Station

Engineers from NASA’s Langley Research Center in Hampton, Virginia, and Boeing dropped a full-scale test article of the company’s CST-100 Starliner into Langley’s 20-foot-deep Hydro Impact Basin. Although the spacecraft is designed to land on land, Boeing is testing the Starliner’s systems in water to ensure astronaut safety in the unlikely event of an emergency during launch or ascent. Testing allows engineers to understand the performance of the spacecraft when it hits the water, how it will right itself and how to handle rescue and recovery operations. The test is part of the qualification phase of testing and evaluation for the Starliner system to ensure it is ready to carry astronauts to and from the International Space Station.

A non-tropical low pressure system that has a possibility of developing subtropical characteristics has developed in the Northern Hemisphere tropics. During the week of January 4, Tropical Storm Pali developed in the Northern Central Pacific Ocean. On January 10 an area of low pressure developed in the Northern Atlantic Ocean, far to the east of Bermuda and NASA's Aqua satellite captured a look at the system in infrared light.

Chemicals: New European Commission determined to make REACH a success

Vice President Tajani and Commissioner Poto?nik have swiftly followed up on their undertaking given during their Hearings in the European Parliament by together visiting the European Chemicals Agency, ECHA, in Helsinki on 25 March. They are taking action to make progress towards the registration of chemicals and the management of substances of very high concern. Of particular importance is the smooth implementation of the registration of high-volume chemicals by the REACH deadline of 30 November 2010 by implementing solutions to practical problems to support industrys work, in particular SMEs. Both Commissioners also announced their common approach on the identification and management of substances of very high concern. They also welcomed progress on the translation of the guidelines for the implementation of REACH. Both commissioners underlined their commitment to the success of REACH, the European Chemicals Regulation.

Broadband : Commission consults on regulatory strategy to promote very high speed Internet

T he European Commission has launched a public consultation on its revised proposals for the regulation of Next Generation Access (NGA) broadband networks, in the form of a draft Commission Recommendation. A previous public consultation held during the last quarter of 2008 confirmed general support for the objective of the Commission to achieve a common regulatory framework for NGA in order to foster timely investment in very high speed networks while ensuring that the competitive structure of the market is maintained. In the light of comments from stakeholders, the revised draft Recommendation includes mechanisms to allocate the investment risk between investors and operators seeking access to NGA networks. The draft Recommendation forms part of the European Broadband Strategy that the March European Council invited the Commission to develop by the end of 2009. The public consultation will be open until 24 July 2009. The Commission plans to adopt the Recommendation, taking account of comments received, before the end of 2009.
